Telomeric chromatin and telomere fragility

Until recently, the prevailing view was that primarily telomere shortening influences ageing and carcinogenesis. However, telomere fragility has now been recognized as another major factor controlling these processes. Telomere fragility is mostly linked to replication stress, which occurs with high propensity at telomeres or sub-telomeric regions due to sequence repeats, specific chromatin structures or DNA topological constraints. By bringing together experts from different disciplines, we aim to discuss the latest developments as well as challenges in our understanding of the structure, function and pathology of telomeres in the context of genome replication and cell differentiation. Specifically, we will focus on mechanisms that either prevent DNA damage as a result of replication stress at telomeres or deal with lesions at telomeric regions in the wider context of the cell cycle. We will also discuss structural aspects of telomeric chromatin and how telomere specific chromatin conformations vary between cell types, e.g. pluripotent and cancer cells. Hence, the topic of the EMBO Workshop is novel, bringing together a unique set of speakers to discuss their work in the following sections: replication stress in general and specifically at telomeres; factors which act at stalled or collapsed replication forks at telomeres; specialized telomeric chromatin structures.
